β-Nicotinamide adenine dinucleotide (NAD+, CAS 53-84-9) is a dinucleotide coenzyme, not a peptide: two nucleotides — one bearing nicotinamide, one bearing adenine — joined through a pair of bridging phosphate groups. It is the central hydride acceptor of cellular redox metabolism, cycling between the oxidised NAD+ and reduced NADH forms in glycolysis, the citric acid cycle and β-oxidation, and it is also consumed as a substrate by sirtuins, PARPs and CD38 rather than merely recycled.
Molecular formula C21H27N7O14P2, molecular weight approximately 663.43 Da. In the laboratory it is used as a cofactor in enzyme assays, as a standard in NAD+/NADH quantification, and as a substrate in sirtuin and PARP activity work. The free acid is hygroscopic and degrades in aqueous solution at neutral to alkaline pH, so it is supplied lyophilised and buffered.
